This book provides an update and detailed review of our knowledge about the newly identified diverse functions of semaphorins, including their potential therapeutic applications. Although semaphorins were originally identified as repulsive axon guidance molecules that were required to direct neuronal axons to their appropriate targets, more than 20 types of semaphorin have since been discovered. Several members of semaphorins, so-called "immune semaphorins," are critical to various phases of the immune response by regulating immune cell-cell contacts or cell migration. In recent years it has also become apparent that semaphorins perform multiple functions in a range of physiological processes, including cardiogenesis, angiogenesis, vasculogenesis, osteoclastogenesis, and immune regulation. In addition, we now know that semaphorins and their receptors can be a potential therapeutic target for various human diseases, including tumorigenesis/tumor metastasis and, neuroregenerative diseases, sudden death and immune disorders.
